Cancers of unknown primary site (CUP) are metastatic tumours for which a standardised diagnostic work-up fails to identify the site of origin at the time of diagnosis. The updated ESMO Clinical Practice Guideline provides information on the definition, incidence and biology, staging and treatment of these cancers, as well as a review of current personalised medicine strategies to identify patients who can expect to benefit from directed therapy.
